چکیده

The study of conformational equilibria and equilibration by nuclear 111agnet£c resonance in cyclic co111pou11ds has been found lo be considerably facilitated by use of 19 F spectra of gemfluoro derivatives. R esults obtained zcith cyclohexane, cis-decali11, and cyclohepta11e ring systems were re·vieu·ed zcith particular emphasis on the degree with which conclusions drawn might also be applicable to the parent cyclic hydrocarbons. This article is based on The Chemical Society Centenary L ecture given by Professor Roberts at Glasgow, Nlanchester, London and East. A11gl£a.

An integrative approach combining ion mobility mass spectrometry, X-ray crystallography, and nuclear magnetic resonance spectroscopy to study the conformational dynamics of α1-antitrypsin upon ligand binding

Native mass spectrometry (MS) methods permit the study of multiple protein species within solution equilibria, whereas ion mobility (IM)-MS can report on conformational behavior of specific states. We used IM-MS to study a conformationally labile protein (α1 -antitrypsin) that undergoes pathological polymerization in the context of point mutations. The folded, native state of the Z-variant rema...

Aspects of the Deter'v1ination of Equilibration Rates by Nmr Spectroscopy

The origins of the line—shape method for determining the rates of chemical and conformational equilibration by NMR spectroscopy are reviewed for the purpose of illustrating the factors which should be considered in obtaining accurate rate data by this very useful technique. Besides a number of illustrations of how changes in rates of conformational equilibration change NMR spectra taken from th...

Structural Dynamics Associated with Intermediate Formation in an Archetypal Conformational Disease

In conformational diseases, native protein conformers convert to pathological intermediates that polymerize. Structural characterization of these key intermediates is challenging. They are unstable and minimally populated in dynamic equilibria that may be perturbed by many analytical techniques. We have characterized a forme fruste deficiency variant of α(1)-antitrypsin (Lys154Asn) that forms p...

Analysis of Cerebrospinal Fluid in Diagnosis of Bacterial Meningitis; Using Nuclear Magnetic Resonance Spectroscopy: A Systematic Review

Background: Analysis of biofluids provides a unique window into the biochemical status of a living organism since the composition of a given biofluid will be modulated according to the level of function of the cells that are intimately concerned with its manufacture and secretion. One of the most successful approaches to biofluid analysis has been the application of NMR spectroscopy. Objective...

Determination of Conformational Equilibria in Proteins Using Residual Dipolar Couplings

In order to carry out their functions, proteins often undergo significant conformational fluctuations that enable them to interact with their partners. The accurate characterization of these motions is key in order to understand the mechanisms by which macromolecular recognition events take place. Nuclear magnetic resonance spectroscopy offers a variety of powerful methods to achieve this resul...
